Restriction and Modification System in Brevibracterium divaricatum II

Abstract
|

|
A type 11 modification enzyme, Bdi I methylase has been isolated from Brevibacterium divaricatum KCTC 8035 by the procedures of ammoniun sulfate fractionation, DEAE - cellulose chromatography and phosphocellulose chromatography.
Bdi. I methylase was a site - specific methylase on Bdi I recognition site, 5´ ATCGAT 3´ which has been proven by the facts that pBR 322 DNA cleaved by Bdi I endonuclease and the plasmid pBDIM 116 containing Bdi I methylase gene were not methylated by Bdi I methylase. And pBR 322 DNA methylated by Bdi I methylase was not cleaved by Bdi I endonuclease. Bdi I methylase was maximally active at pH 7.5 and in the presence of l 0mM NaCl.
